The free energy of thiol ester hydrolysis.

چکیده
Thiol esters, such as aeetyl coenzyme A, the principal biological carrier of activated acetyl groups, have generally been supposed to be “energy-rich” compounds; i.e. to have a free energy of hydrolysis at pH 7.0 in the range of -7000 to -14,000 calories per mole. The reported numerical values for the free energy of thiol ester hydrolysis have varied over a wide range (for recent estimates see references 1 and 2) because of changes in the estimates for the equilibria and free energies of formation on which they were based, and Wadso, who has recently measured their heats of hydrolysis, has questioned whether thiol esters are “energy-rich” compounds at all (3, 4). Thiol esters which contain a hydroxyl group in a suitable position for intramolecular attack on the acyl group have been shown by Harding and Owen to isomerize to the oxygen ester in dilute alkali at a faster rate than they undergo hydrolysis (5, 6) :

First-principle studies of intermolecular and intramolecular catalysis of protonated cocaine
We have performed a series of first-principles electronic structure calculations to examine the reaction pathways and the corresponding free energy barriers for the ester hydrolysis of protonated cocaine in its chair and boat conformations. The calculated free energy barriers for the benzoyl ester hydrolysis of protonated chair cocaine are close to the corresponding barriers calculated for the ...
متن کاملInvestigations of thiol-modified phenol derivatives for the use in thiol–ene photopolymerizations
Thiol-ene photopolymerizations gain a growing interest in academic research. Coatings and dental restoratives are interesting applications for thiol-ene photopolymerizations due to their unique features. In most studies the relative flexible and hydrophilic ester derivative, namely pentaerythritoltetra(3-mercaptopropionate) (PETMP), is investigated as the thiol component. Thus, in the present s...
متن کاملHydrolysis of Several Substituted Methyl Benzoates in the Aqueous Solution
-The hydrolysis of several ring substituted methyl benzoates was investigated in aqueous solution over a pH range of 3-10. Both the pH dependence (at pH > 5) and the variation of hydrolysis rates with ring substitution are consistent with a mechanism involving the addition of hydroxide ion to the ester carbonyl group. Extrapolation of the rate data to a pH and temperature that would be consiste...
متن کاملPreparation of acrylic-type derivative of ibuprofen and in vitro evaluation studies of its polymeric prodrugs
Acrylic-type polymeric systems having degradable ester bonds linked to ibuprofen were synthesized and evaluated as materials for drug delivery. Ibuprofen, as a non-steroidal anti-inflammatory drug, was linked to 2-hydroxyethyl methacrylate by activated ester methodology in one-pot procedure. The resulting methacrylic derivative of ibuprofen was copolymerized with 2-hydroxyethyl methacrylate and...
متن کاملUnexpected Reaction Pathway for butyrylcholinesterase-catalyzed inactivation of “hunger hormone” ghrelin
Extensive computational modeling and simulations have been carried out, in the present study, to uncover the fundamental reaction pathway for butyrylcholinesterase (BChE)-catalyzed hydrolysis of ghrelin, demonstrating that the acylation process of BChE-catalyzed hydrolysis of ghrelin follows an unprecedented single-step reaction pathway and the single-step acylation process is rate-determining....
